Kilise 69-155 yılları arasında yaşamış, Aziz Yuhanna’nın ilk öğrencilerinden olan ve sonradan Aziz ilan edilmiş olan İzmir Piskoposu St. Polikarp’a ithaf edilmiştir. Romalı yöneticilerin inancından vazgeçirmek için çok baskı yapmalarına rağmen Polikarp inancından vazgeçmemiş bunun üzerine 23 Şubat 155’de Kadifekale eteklerindeki Roma Stadyumu’nda yakılarak öldürülmüştür. Adı verilen bugünkü kilise 1625 yılında Sultan Süleyman’ın izni ile yapılmıştır. 1680 deki büyük yangından sonra bir onarım görmüştür.Kilise 1775 yılında Osmanlı makamlarının izni ve 16.
