1872 yılında yıktırılan kale arkasında bugünkü Kemeraltı çarşısında, Hisarönü mevkiindedir. 1592 yılında Yakup Bey tarafından yaptırılmıştır. İzmir’in en büyük ve gösterişli camiidir. Yenilenen minaresi ile cami, klasik dönem Osmanlı Camilerinin tipik özelliğini taşımaktadır. Cami içindeki kalem işi süslemeleri, hat sanatının inceliklerinin sergilendiği panolar ve mihrap - mimber işçiliği Türk sanat zevkinin en iyi örneklerindendir. Cami 1813, 1881,1927 ve 1980’li yıllarda onarım görmüştür.
