1900'lü yılların başlarında (1898-1913) Banker Kostaki Teophylaktos tarafından konut olarak yaptırılmıştır. Kostaki Teophylaktos 1917 yılında iflas edince, bu yapıyla birlikte bütün mal varlığına haciz konulmuş ve konak Nemlioğlu ailesi tarafından satın alınmıştır. 15-17 Eylül 1924 tarihinde Trabzon'u ilk ziyaretinde Atatürk, eşi Latife Hanım ve beraberindeki heyetle birlikte bu konakta misafir edilmişlerdir. Ülkemizin sayılı sivil mimarlık örnekleri arasında yer alan konağın bodrum kat hariç tüm kat duvarları tamamen kalem işi süslüdür. Konağın bodrum katı Arkeolojik Eserler Seksiyonu, zemin katı Konak Teşhiri, birinci katı Etnoğrafik Eserler Seksiyonu ve asma katı İdari Bölüm olarak düzenlenmiştir.
