Kanuni Sultan Süleyman zamanında on beş sene sadrazamlık yapan eski Osmanlı vezirlerinden Rüstem Paşa’nın adını taşıyan camii, 1550-1560 yılları arasında Mimar Sinan tarafından yapılmıştır. Mimar Sinan’ın, Sadrazam Rüstem Paşa için, Sapanca’da inşa ettiği camii, imaret, hamam ve kervansaraydan oluşan külliye, XVI. yüzyılın günümüze gelmeyen önemli eserlerindendir. Camii zamanla değişikliğe uğramış ve Rumi 1146’da ikinci defa inşa edilmiştir. Rüstempaşa Camii’nin üstü ahşap çatı ve kiremit ile örtülüdür. Camiinin yan cephesindeki yeni yazılı kitabede “Rüstempaşa Camii Yapım Tarihi 1554” ibaresi bulunmaktadır. Camiinin duvarları alçaktır. Kerpiç-ahşap karışımı bir binadır. Camiinin toplam arsa alanı 750 metrekare olup, iç alanı 250 metrekaredir ve kapasitesi 1.200 kişidir.
