Gaziantep Kalesi, ilk olarak Roma Dönemi'nde höyük üzerinde bir gözetleme kulesi olarak yapılmış, bugünkü biçimini ise “Kaleler Mimarı” olarak adlandırılan Bizans İmparatoru Justinyanus döneminde MS. VI. yüzyılda almıştır. Kale çapı yaklaşık 100 metre, çevresi 1200 metre olan gayrı muntazam dairesel bir şekle sahiptir. Kale bedenleri üzerinde 12 adet kule mevcuttur.
