Ulu Camii’nin kuzeyinde ve camiye bitişik olan Mesudiye Medresesi Diyarbakır’da yapılan ilk büyük medresedir. Medresenin yazıtından öğrenildiğine göre 1198-1199 yılında Artuklu Meliki Mesud Kutbeddin Ebu Muzaffer Sökman zamanında yapılmaya başlanmış, Sökmen II'nin ölümünden sonra yerine geçen Mesud zamanında yapım çalışmaları devam etmiş, Mevdud zamanında 1223’de tamamlanmıştır Medresenin mihrap yakınındaki pencerelerden birisi üzerindeki kitabeden de planlarını Halepli usta Cafer İbni Mahmut ‘un çizdiği ve yapımını da Mesud’un sürdürdüğü öğrenilmiştir.
