Cami, 1534-1537 yılları arasında Diyarbakır Valisi Hadım Ali Paşa tarafından yaptırılmıştır. Cami Mimar Sinan’ın Diyarbakır’daki eserlerinden biridir. Bugün kullanılmayan Şafiler bölümü 1769-1770 yılları arasında eklenmiştir. Ayrıca külliyede Ali Paşa Medresesi, zikirhane, hamam ve avlunun ortasında bir şadırvan yer almaktadır.
